Scientists have identified a previously underappreciated region of the brain that may explain how medications like Ozempic reduce cravings for food, alcohol, and other addictive substances.1 The lateral septum, a brain area densely populated with GLP-1 receptors, acts as a control hub between thinking about rewards and pursuing them.1 Researchers at the University of Otago, led by Robert Munn and his laboratory, discovered and validated that GLP-1 agonists—drugs that mimic the GLP-1 hormone—work by directly activating this region to suppress consumption.1
GLP-1 agonists including Ozempic and Wegovy stimulate insulin release, slow digestion, and increase feelings of fullness.1 Clinical evidence shows these medications reduce alcohol consumption in humans, while animal studies demonstrate they can decrease use of cocaine, amphetamine, opioids, and nicotine.1 The lateral septum receives most of its primary input from the hippocampus and, together with that region, processes information about location and timing.1 Significantly, this area contains place cells that respond strongly to rewards, effectively adding information about "what is good in this location" to spatial and temporal data.1 Direct activation of GLP-1 in the lateral septum reduced both food and alcohol consumption in mice during testing.1
