重建后的皇岗口岸于10月12日上午6时30分正式开通运行，提供24小时通关服务123。开通首日从6时30分至24时，共有7.95万人次出入境，其中入境4.13万人次、出境3.82万人次；出入境车辆达0.8万辆次，其中入境0.42万辆次、出境0.38万辆次3。

首位入境的私家车主谢先生于6时45分通过口岸，仅用几分钟就完成通关1。首位出境旅客、深圳旅客陈先生对新建的超级口岸联检大楼印象深刻，表示通关效率很高、流程顺畅2。
