b.next, a company conducting academic research in synthetic cell development, established a manufacturing facility to produce Cytosol, a core material used in synthetic cells, after encountering critical supply chain disruptions.1 The company faced constraints when Roche halted tRNA supplies in 2023, followed by Sigma's discontinuation of related reagents.1
To address these challenges, b.next undertook more than two years of development work on an open-source PURE system, overcoming material licensing restrictions and reagent supply interruptions.1 Beginning in mid-2024, the company shared PURE protocols and plasmids through its Nucleus platform, and by mid-2025 had completed open-source protocols and specifications for all PURE components.1 The facility in San Francisco became operational in late 2025 to early 2026.1
Since establishing operations, b.next has supplied over one hundred Cytosol reagent kits to approximately twenty laboratories worldwide over the past six months.1
